The optimized formulation sinks more than three times as effectively as conventional loading buffers, reducing cross-well contamination. A new reducing agent efficiently reduces disulfide bonds without the toxicity and odor of DTT or β-mercaptoethanol. Mix buffer and sample at 1:4 (5X) or 1:1 (2X), heat at 95°C, briefly centrifuge and load. SDS gives proteins a dominant negative charge, reducing differences in intrinsic charge. Bromophenol blue tracks electrophoresis progress.
